    题名: FADS2 polymorphisms correlate with HDL-C and desaturase activity in type 2 diabetes

    摘要: Genetic variations of fatty acid desaturase (FADS) genes, encoding rate-limiting enzymes for PUFA synthesis, have been reported to correlate with blood lipids levels and desaturase activities mostly in Caucasians healthy populations. We aimed to examine whether the FADS single nucleotide polymorphisms (SNPs) would correlate with blood lipid levels and FADS activities in type 2 diabetes in Taiwan. A total of 874 patients were recruited from the Diabetes Management through Integrated Delivery System (DMIDS) cohort at 2008. Of which, 173 patients without using lowering lipid drugs were selected in this investigation. FADS2 SNP rs174576 to serve as the tag SNP of rs174548, rs174550, rs174583 and rs2727270 was genotyped, and these SNPs were in strong linkage disequilibrium (LD) (r2蠅0.8).Tag SNP rs174576 C allele was associated with higher levels of HDL-C, but not cholesterol,triglyceride,non-HDL-C and LDL-C concentrations. The 174576 C allele carriers also showed higher Δ5 (20:4n-6/20:3n-6) and Δ6 (18:3n-6/18:2n-6) fatty acid desaturase activities. Other SNPs including rs174548, rs174550, rs174583 and rs2727270 in the same LD block also showed similar results. FADS2 SNP of this LD block was associated with higher HDL-C concentrations as well as andΔ5 andΔ6 desaturase activities. It will be worth exploring whether genetic variations of FADS2 would exert effects on diabetic cardiovascular complications via modifying HDL-C and desaturase activities.
