    題名: Ethanol metabolizing genes and risk of head and neck cancer: Preliminary report

    摘要: Head and neck cancer (HNC), including cancers of theoral cavity, pharynx, and larynx, is the fifth most com-mon cancer in the world. One of the major risk factorsof HNC is alcohol drinking; however, most drinkers donot develop HNC, suggesting a role of genetics. Thisanalysis examines the association between HNC risk andtwo functional single nucleotide polymorphisms (SNPs)of two ethanol metabolizing genes (ADH1B: rs1229984,Arg48His and ALDH2: rs671, Glu504Lys). HNC patientsand frequency-matched (by age and sex) non-cancer con-trols were recruited from the department of otolaryn-gology and department of stomatology. Information onalcohol consumption was ascertained by interviews. Geno-typing was performed using TaqMan based real-time PCR.Unconditional logistic regression was performed with dataof 88 HNC cases and 81 controls to estimate HNC riskassociated with the two SNPs adjusted for age and sex.The results showed that those who carried at least oneADH1B 48Arg allele were twice as likely to develop HNCcompared to homozygous 48His individuals [Odds ratio(OR)=1.97; 95% con?dence interval (CI): 1.04-3.72]. Thepositive association between ADH1B 48His and HNC riskwas more prominent among those who were heterozygousfor ALDH2 Glu504Lys (OR=3.19; 95% CI: 1.17-8.74) andamong never drinkers (OR=5.51; 95% CI: 1.32-23.06). Thisstudy shows a significant contribution of ADH1B Arg48Histo the development of HNC, which is modified by ALDH2Glu504Lys and alcohol drinking.
