    Title: Analysis of astrocytoma heterogeneity: Detection of telomere shortening, BCL-2 expression, and multiple gains and losses of genetIc material in four primary cultures of astrocytoma

    Abstract: For understanding of the biological heterogeneity of human astrocytoma, we previously established four primary cultures (designated as #922, 901, 908 and 831) from the tumor specimens of four astrocytoma patients. They were all staining positive for astrocyte-specific intermediate filament protein GFAR, and exhibited activity in Matrigel invasion assay. In this study, we analyzed their genetic aberrations via comparative genomic hybridization (CGH), and found that chromosome loss occurred in only one case, i.e. 19p13.1-13.2 in#901; however, the chromosome gains occurred in two of four primary cultures include 5q35, 7p22, 9q34, 13q33, 16p13.3, 16q24, 17q25, 19p13.2-ter, 21q22 and 22q13. In addition, the telomere length and Bcl-2 expression level were also investigated in these four primary cultures. The order of telomeric length average, #831>908>922>901, was reverse correlated with the cell growth rate. Interestingly, the order of Bcl-2 expression level was reverse correlated with the order of telomeric length average, i.e. #901>922>908>831. Our data suggest that Bcl-2 overexpression could rescue the harmfulness of telomere shortening caused by elevated proliferation during malignant progression of astrocytoma.
