Title: | DNA methylation at birth and IgE trajectories through childhood |

Abstract: | Rationale: Prenatal exposure can modulate offspring DNA methylation (DNAm) at birth. The association of such DNAm with IgE trajectory later in life and whether such association is race-specific are unclear. Methods: We examined two independent birth cohorts, the Isle of Wight birth cohort (IOWBC) in UK (n=796; all White) and a birth cohort in Taiwan (n=60; all Asian). For both cohorts, genome-wide DNAm at birth and IgE at multiple ages since birth were analyzed. Semi-parametric regressions via PROC TRAJ in SAS were used to detect IgE trajectories. Using the R package ttSCreening, we tested the association of DNAm with the trajectories using linear regressions with randomly selected training/testing data to effectively control for multiple testing. Logistic regressions were further applied to include covariates cell type compositions and sex. Biological pathways and methQTLs for the identified CpGs were studied for both cohorts. Results: In total, 103 CpGs were identified in IOWBC and 476 CpGs in the Taiwan cohort, of which only one CpG was in common, cg16711274 (MINAR1), an methQTL site. Although minimal overlap in CpGs, further assessment on biological pathways indicated that 17 pathways were common between the two cohorts, of which at least four are linked to airway diseases, e.g., glycerophospholipid metabolism is relevant to asthma pathogenesis. Conclusions: DNAm at birth at multi-sites is associated with IgE trajectory later in life, and CpGs showing such associations are likely to be different between White and Asian, although the CpGs may be enriched in the same pathways linked to airway diseases. |
