    題名: The regulatory role of Sphingosine-1-Phosphate (S1P) in the development of airway hyperresponsiveness

    摘要: Rationale: Airway hyperresponsiveness to bronchoconstrictor stimuli is a key feature of asthma. S1P is a bioactive sphingolipid that plays important roles in allergic responses, including asthma. Our study was to investigate the association between airway inflammation and S1P, in individuals with asthma and pulmonary epithelial cells as a model in vitro. Methods: Various cells were stimulated for airway contraction and inflammatory assay. The levels of S1P were evaluated for their relationship with pulmonary function tests. Results: In a case-control study, we found that the plasma level of S1P was significantly higher in 80 asthma cases. In asthmatic children there was significant positive correlation between the levels of S1P and eosinophils and ECP. Investigating the association of bronchial hyper-responsiveness with S1P, our studies showed that higher S1P levels clearly led to positive results more swiftly. It was found that S1P caused a potent contraction of ASMC (airway smooth muscle cell)-embedded collagen gel, and the gel surface area was reduced by 55% versus that of the control by 33%. Pretreatment of cells with S1PR2 antagonist (JTE013) obviously blocked S1P-induced ASMC contraction. Moreover, by comparing PBMC between asthma patients and healthy controls, we found that the levels of S1PL (S1P Lyase) activity of controls were higher. In the study of lung epithelial cell showed that S1P was able to induce, in a dose-dependent manner, the expression of MCP-1 primary bronchial epithelial cells. Conclusions: Under stress conditions, generation of S1P in the lung, particularly the epithelial cells, may contribute to the increased airway hyperresponsiveness and inflammation.
