    題名: Knocking out of SWAP-70 makes spontaneously transformed NIH3T3 cells normal

    摘要: Recently we showed that SWAP‐70 knocked out mouse embryo fibroblasts (MEFs) did not transform for a long time and adding back of SWAP‐70 restored transforming ability. It is well known that NIH3T3 cells easily transform spontaneously. This year we used spontaneously transformed NIH3T3 cells. Using a kit, the SWAP‐70 gene was knocked out. After knocking out of the SWAP‐70 gene, growth rate of the cells was diminished and contact inhibition ability was restored. They required high concentration of serum to grow whereas the spontaneously transformed cells grew in the medium with low concentration of serum. They became resistant to nutrient depletion when the spontaneously transformed cells were sensitive to nutrient depletion. The parental the spontaneously transformed NIH3T3 cells grew in soft agar but the SWAP‐70 knocked out cells did not. Finally, the spontaneously transformed cells made tumors in nude mice but the SWAP‐70 knocked out cells did not. Taken together, SWAP‐70 is required for spontaneous transformation of rodent cells. It is interesting to see what happens when the SWAP‐70 gene of human tumors is disrupted.
